Flyspeck products are built to work with your EXISTING site templates and content. Drupal and Mambo are more "site frameworks" that take over your whole site, your site is really built "in" them, not "around" them. These systems are great, but customization is difficult, and it can be overkill for many sites. They also require a database, which adds additional technical know-how and maintenance.
